Kinetics of Leukemic

چکیده
My associates Drs. Frank Schabel and William Wilcox and I recently finished writing a monograph on the criteria and kinetics associated with “curability― of experimental leukemia (7). That lengthy manuscript, which is now “in press,―covered in detail' some 8 years of study on the very subject I have been asked to discuss today. We are happy to report that the work we have done since all that writing does not provide the basis for another long paper devoted to retractions or conflicting results and con. clusion.s. l\/Iy dilemma should be apparent ! Most of what is new that we have learned about the kinetics of leukemic cell behavior is “in press―and hence has not yet become avail able for consideration by the participants of this con ference. Neither I nor the sponsors of this conference wish to be a party to repetitious publication. However, I must either begin my discussion in the middle or be allowed to restate briefly a few background observations (and correllary deductions) that already have been made and documented (7): “1. One leukemic cell is lethal ; if one leukemic cell sur vives therapy, it may proliferate to a number which will kill the host. Therefore, since cure, not just temporary remission or increase in host life span is the goal—a most important guide line or value regarding the response of leukemia to therapy is the number of leukemic cells which survive treatment; i.e., each given dosage level and any given schedule and including combinations of agents ad ministered simultaneously or sequentially. “2. The dynamics of the proliferation of L1210 leukemic cells as employed herein may be described as follows:

Spectral discrimination between normal and leukemic human sera using delayed luminescence
In this work, photoinduced delayed luminescence (DL) was used to distinguish serum samples of patients with acute lymphoblastic leukemia from those of healthy volunteers. DL decay kinetics of human serum samples was measured using a homebuilt ultraweak luminescence detection system. It was found a significant difference in the weight distribution of the decay rate between normal and leukemic se...

Spleen colony assays were used to quantitate the cytocidal effect of actinomycin D on leukemic cells and normal hematopoietic stem cells. Different doses of actinomycin D were administered to either normal or leukemia-bearing mice, and the surviving fraction of these cell populations was determined. Following single injections of the drug, the killing of both the normal and leukemic cell popula...

متن کاملKinetics of normal hematopoietic stem and progenitor cells in a Notch1-induced leukemia model.
The predominant outgrowth of malignant cells over their normal counterparts in a given tissue is a shared feature for all types of cancer. However, the impact of a cancer environment on normal tissue stem and progenitor cells has not been thoroughly investigated. We began to address this important issue by studying the kinetics and functions of hematopoietic stem and progenitor cells in mice wi...
